Cyanoacetamide and ethyl acetoacetate undergo condensation cyclization, chlorination, cyanohydrolysis, Hoffman degradation, catalytic hydrogen release of chlorine, selective chlorination to produce 2-chloro-3-amino-4-methylpyridine, and then undergo amidation with 2-chloronicotinyl chloride, cyclopropylamine nucleophilic substitution and closed-loop reaction to generate nevirapine, with a total yield of 39%.
